Melatonin — 1 mg/ml
Melatonin is the body’s darkness signal. It binds MT1/MT2 receptors in the brain’s clock and helps set “night has started.” Low-dose melatonin (around 0.3–1 mg) is commonly used to support sleep onset and circadian timing without the next-day grogginess often linked to much higher doses.
GABA — 100 mg/ml
GABA is the main inhibitory neurotransmitter. Supplemental GABA is used to support a calmer pre-sleep state and shorter time to fall asleep; typical study doses sit in the 100–300 mg range. Effects may be partly peripheral (gut–vagus) as well as central. Glutamine in the same formula can feed the body’s own GABA pathway.
L-Arginine — 100 mg/ml
Arginine is the precursor for nitric oxide (NO). NO signalling in sleep-related brain regions has been linked to more slow-wave sleep and a more relaxed, vasodilated state. It also supports overnight recovery and circulation.
L-Glutamine — 25 mg/ml
Glutamine is a precursor for glutamate and then GABA, so it supports the same calming pathway as the added GABA. It is also a primary fuel for the gut lining and immune cells during the night, when recovery and nitrogen balance matter most. Similar night blends use ~25–30 mg/ml glutamine for that dual role.
Why the four together
The formula hits several overnight levers at once:
• clock / “it’s night” (melatonin)
• quiet the nervous system (GABA + glutamine → GABA)
• NO / relaxation and sleep architecture (arginine)
• gut, immune and recovery support while you sleep (glutamine)
A commercial analogue uses the same GABA + arginine backbone with slightly higher melatonin and glutamine, described as targeting circadian, GABAergic, nitric-oxide and nocturnal recovery pathways together
