Epitalon (also spelled Epithalon or Epithalone) is a synthetic four-amino-acid peptide with the sequence alanine-glutamic acid-aspartic acid-glycine (Ala-Glu-Asp-Gly or AEDG). It was created by Professor Vladimir Khavinson and colleagues at the St. Petersburg Institute of Bioregulation and Gerontology as a pure, defined analog of Epithalamin, a polypeptide complex originally extracted from bovine pineal glands. Unlike the complex natural extract, Epitalon is a single, precisely sequenced tetrapeptide, offering consistency, purity, and freedom from animal-source variability.

Its most studied action is the activation of telomerase. In human somatic cell cultures, Epitalon has been shown to induce expression of the catalytic subunit of telomerase (hTERT), restore enzymatic telomerase activity, and produce measurable elongation of telomeres. In classic experiments, treated human fibroblasts continued dividing well beyond the normal Hayflick limit while maintaining youthful morphology and telomere length. This telomere-maintenance effect is central to its reputation as a geroprotective (anti-aging) research compound.

Beyond telomeres, Epitalon influences the pineal gland. It upregulates the rate-limiting enzyme in melatonin biosynthesis (AANAT), helping restore the natural nighttime melatonin peak that declines with age. This supports healthier circadian rhythms, improved sleep quality, and downstream antioxidant and immune benefits. Additional reported actions include epigenetic modulation of gene expression, reduction of oxidative stress, normalization of certain immune markers, and mild neuroprotective and anti-mutagenic effects observed in animal models.

Research—primarily from Russian laboratories over more than two decades, with some independent confirmation in recent years—has linked Epitalon to extended cellular replicative lifespan, improved melatonin secretion in aged subjects, reduced chromosomal aberrations, and lifespan or healthspan benefits in rodent studies. Human data exist but remain more limited in scale and independent replication compared with many mainstream pharmaceuticals.

In practical research and wellness contexts, Epitalon is valued for addressing multiple hallmarks of aging simultaneously: telomere attrition, circadian disruption, oxidative stress, and declining pineal function. Short cyclic protocols are commonly used because the peptide appears to produce lasting changes in gene expression and cellular behavior rather than requiring continuous daily administration.

Epitalon therefore stands as a compact, multi-pathway pineal bioregulator focused on cellular longevity, melatonin restoration, and healthy aging research.
