5-Amino-1MQ (5-Amino-1-methylquinolinium, sometimes shortened to 5-Amino-1Q) is a lab-made synthetic small molecule – NOT a peptide or hormone -that works as a potent inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT). Developed in the mid-2010s through structure-guided design, it is taken orally as capsules and targets a key metabolic ‘brake’ that becomes overactive in obesity and aging. By blocking NNMT, it helps your cells hold onto more NAD+ (the energy coenzyme we discussed with NAD+ injections) and shifts metabolism toward fat burning and efficiency.
Think of 5-Amino-1MQ as a cellular ‘NAD+ guardian and fat-burn accelerator.’ It removes a metabolic bottleneck that wastes energy and promotes fat storage.
How it works (super simple):
– Blocks NNMT (overactive in obese fat tissue) -prevents nicotinamide from being ‘thrown away’ as 1-MNA.
– This preserves and boosts intracellular NAD+ and SAM levels – supercharges mitochondria, ramps up fat oxidation, shrinks fat cells, and improves insulin signaling.
In short: It makes your cells act like they’re on a fat-burning, high-energy program – similar to exercise or NAD+ boosting but through a unique NNMT pathway.
Preclinical mouse and cell studies show an excellent short-term safety profile – no observable adverse effects even at effective doses.
In available data: No significant toxicity, no changes in food intake, no organ damage reported.
Mild/rare effects (mostly anecdotal from clinic users):
– Temporary digestive changes or mild headaches (often tied to hydration/electrolytes in first week).
– Occasional muscle soreness as metabolism ramps up.
– Theoretical risks unproven (e.g., long-term NAD+ pathway shifts).
Bottom line: Looks very well-tolerated in research settings, but without human data this is still pure experimental territory.
There are no official guidelines (never tested in humans), so these come from mouse scaling, preclinical papers, and anecdotal clinic protocols.
Method of Administration: Subcutaneous into fatty tissue.
Reconstitution: 2ml Bac water into single vial. See Reconstitution Guide
Anecdotal Dose: Start low 2mg daily, therefore @2mg daily, 40 units or 0.4ml. Use peptide calculator for alternative dosage. Many researchers increase dosage as and when necessary.
Course: 5 days @ 2mg.
Cycling: Use whilst on weight-loss drugs to keep muscle. 4 weeks on/4weeks off. See article.
Tip: Reconstitute with bacteriostatic water; store in fridge. Use insulin syringes and sterile technique.
– Fat-cell shrinker: it literally makes individual fat cells smaller (3040% in studies) rather than just emptying them.
– NAD+ saver: instead of adding NAD+ directly (like the injections we covered), it stops your cells from wasting the precursor: a smarter long-term boost.
– Exercise mimetic cousin: works alongside compounds like SLU-PP-332 or MOTS-c for metabolic upgrades without the gym.
– Obesity enzyme target: NNMT is dramatically overexpressed in fat tissue of overweight people; inhibiting it ‘normalizes’ metabolism.
– Rapid mouse results: obese mice dropped measurable weight in under 2 weeks with zero change in appetite.
– Not a peptide: tiny chemical (no amino-acid chain), which is why it is stable as easy capsules unlike most peptides.
– Aging research star: higher NNMT activity speeds up aging; blocking it is now a hot longevity target.

Takeaway: Impressive mouse proof-of-concept for fat loss and metabolism, but we desperately need human trials.
