PE-22-28 is a lab-made synthetic heptapeptide – a tiny chain of just 7 amino acids derived as an optimized C-terminal fragment (positions 22-28) of the naturally occurring Spadin peptide (itself a cleavage product of sortilin/propeptide). Developed through research on TWIK-related K+ channel 1 (TREK-1) modulation, it represents a novel approach to mood disorders and cognitive enhancement. It is administered in research settings as a nasal spray or subcutaneous injection and has gained attention for its rapid antidepressant-like effects without the side effects or delayed onset of traditional SSRIs.
Think of PE-22-28 as a ‘fast-acting brain reset button.’ It selectively blocks a specific potassium channel to quickly boost neuronal activity and repair pathways.
How it works:
-Potent and selective antagonist of TREK-1 potassium channels.
-Causes neuronal depolarization – increases serotonergic neuron firing in the dorsal raphe nucleus – rapid BDNF upregulation, TrkB receptor sensitivity, and enhanced neuroplasticity/neurogenesis (new neuron formation and connections).
– Rapid antidepressant effects: onset in days (vs weeks for SSRIs); reduces depression-like behaviors in forced swim and novelty-suppressed feeding tests.
– Cognitive enhancement & memory support: improves learning, executive function, and hippocampal neurogenesis.
– Neuroprotection & stroke recovery: protects against ischemia, prevents post-stroke depression, and supports motor/cognitive recovery in models.
– Bonus signals: potential anti-anxiety effects, improved stress resilience, and modulation of inflammation without broad immunosuppression.
In short: It delivers fast mood, cognitive, and neuroprotective benefits by flipping the brain’s excitability switch – a promising alternative to traditional antidepressants in research.
PE-22-28 shows an excellent safety profile in preclinical studies – no notable toxicity or major side effects even at effective doses. In available data: Extremely well-tolerated; effects are localized to brain pathways without systemic hormonal disruption.
Mild/rare effects:
– Temporary headache, lethargy, or mild nasal irritation (with spray).
– No reports of serotonin syndrome, sedation, or dependence.
Bottom line: One of the gentlest nootropic/antidepressant peptides in research – side effects are minimal and far milder than traditional medications.
Method of Administration: Subcutaneous into fatty tissue.
Reconstitution: 2ml Bac water into single vial. See Reconstitution Guide
Dose: 250mcg daily, therefore 5 units or 0.05ml. Use peptide calculator for alternative dosage.
Course: 4-8 weeks on. 2-4 weeks off.
Cycling: Repeat every 3 months for maintenance or as needed (e.g., during respiratory seasons or after illness). Many protocols use it intermittently during high-stress periods or short courses for neuroprotection.
Tip: Reconstitute with bacteriostatic water; store in fridge. Use insulin syringes and sterile technique.
Human data is extremely limited (mostly preclinical mouse/in vitro); strong mechanistic evidence from Spadin analogs.

8. PEP DOSE dosage calculator (2026): Optimal 400 mcg nasal once daily (morning); 400600 mcg for maintenance in depression protocols.
9. ScienceDirect / Pietri stroke study (2019, referenced 2026): Low-dose activation vs higher-dose inhibition of TREK-1 for biphasic neuroprotective/antidepressant effects.
10. Peptide Sciences & comparative overviews (2026): Stronger potency/stability than parent Spadin; no QT prolongation or cardiac side effects in models.
Takeaway: Promising preclinical data for rapid mood/cognitive benefits; human trials are needed for validation.
